The effect of the bacterial cytolytic toxin, streptolysin S, on liposomes composed of various phospholipids was investigated. Large unilamellar vesicles containing [14C]sucrose were prepared by reverse-phase evaporation, and membrane damage produced by the toxin was measured by following the release of labeled marker. The net charge of the liposomes had little or no effect on their susceptibility to steptolysin S and the toxin was about equally effective on liposomes composed of phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine and phosphatidylglycerol. Experiments with liposomes composed of synthetic phospholipids showed that the ability of the toxin to produce membrane damage depended on the degree of unsaturation of the fatty acyl chains. The order of sensitivity was C18 : 2 phosphatidylcholine > C18 : 1 phosphatidylcholine > C18 : 0 phosphatidylcholine = C16 : 0 phosphatidylcholine. Liposomes containing the latter two phospholipids were virtually unaffected by streptolysin S, and experiments with C18 : 0 phosphatidylcholine suggested that toxin activity does not bind to liposomes composed of phospholipids with saturated fatty acyl chains. The inclusion of 40 mol% cholesterol in C16 : 0 phosphatidylcholine and C18 : 0 phosphatidylcholine liposomes made these vesicles sensitive to streptolysin S. Egg phosphatidylcholine liposomes, which were unaffected at 0°C and 4°C became susceptible to the toxin at these temperatures when cholesterol was included. Liposomes composed of C14 : 0 phosphatidylcholine were unaffected by streptolysin S at temperatures below the chain-melting transition temperature (23°C) of this phospholipid, but became increasingly susceptible above this temperature. The results suggest that the fluidity of the phospholipid hydrocarbon chains in the membrane is important in streptolysin S action.  相似文献

Epicuticular wax of Cirsium arvense contains hydrocarbons (12%), esters (35%), free acids (3%), free alcohols (10%), triterpene acetates (8%) and 1,3-ditetradecanoyl-2-hexanoylglycerol (8%) as major components. Natural and human‐made disasters such as floods and logging occur in and around rivers. Stream‐dwelling aquatic insects respond to these disturbances in various ways. Primary consumers among them rely greatly on algae and leaf litter from riparian vegetation as food. Therefore, once a disturbance such as a flood has occurred, insects may find it difficult to find food in a stream, and the aquatic insect assemblage can be impacted greatly as a result. Disturbances in riparian areas also increase fine sediment loads into streams, damaging habitat and altering the aquatic insect assemblage. Deforestation impacts not only terrestrial but also aquatic animals. In this review paper, aquatic insect assemblages are assessed according to alterations in land use in and around streams. Following this paper, it is expected that clarification of aquatic insect fauna and their life cycles will progress and that the distribution and habitat use of aquatic insects will be afforded greater attention in forest management.  相似文献

Little is known about the potential activity of microbial communities in hypersaline sediment ecosystems. Ribosomal tag libraries of DNA and RNA extracted from the sediment of Lake Strawbridge (Western Australia) revealed bacterial and archaeal operational taxonomic units (OTUs) with high RNA/DNA ratios providing evidence for the presence of ‘rare’ but potentially “active” taxa. Among the ‘rare’ bacterial taxa Halomonas, Salinivibrio and Idiomarina showed the highest protein synthesis potential. Rare but ‘active’ archaeal OTUs were related to the KTK 4A cluster and the Marine-Benthic-Groups B and D. We present the first molecular analysis of the microbial diversity and protein synthesis potential of rare microbial taxa in a hypersaline sediment ecosystem.  相似文献

The field of gerontology is remarkably diverse; yet there has been relatively little investigation of physical anthropological issues in aging research. This review explores gerontologic topics of actual and/or potential interest to physical anthropologists. The evolution of aging presents a theoretical dilemma in that postreproductively expressed traits may be outside the influence of natural selection. The physiological changes of aging comprise a diversified mosaic of physical deterioration as would be expected from an evolutionary model. Studies of prehistoric aging are limited to estimating lifespan, which may not be indicative of rate of aging. Considerable attention has been devoted to body composition and aging, and notable findings include a loss of lean tissue with age and relatively constant (though redistributing) fat mass. Osteoporosis is a major problem of aging in females, as is tooth loss in both sexes. The study of variation in rates of aging is only beginning, and a number of approaches to measuring biological age in adulthood are presented. Determining the associations of lifestyle, economic, and nutritional status with biological age may reveal sources of variation in rates of aging and may be of practical importance.  相似文献

Fungi impact humans and the environment in many ways, for good and ill. Some fungi support the growth of terrestrial plants or are used in biotechnology, and yet others are established or emerging pathogens. In some cases, the same organism may play different roles depending on the context or the circumstance. A better understanding of the relationship between fungal biochemical composition as related to the fungal growth environment is essential if we are to support or control their activities. Synchrotron FTIR (sFTIR) spectromicroscopy of fungal hyphae is a major new tool for exploring cell composition at a high spatial resolution. Brilliant synchrotron light is essential for this analysis due to the small size of fungal hyphae. sFTIR biochemical characterization of subcellular variation in hyphal composition will allow detailed exploration of fungal responses to experimental treatments and to environmental factors.  相似文献

The surgical castration of male chickens induces hormonal changes, which permanently influence metabolic processes in birds. The aim of this study was to determine the effect of age and castration on the growth rate, feed conversion, lipid profile and histopathological changes in the livers of cockerels and capons. The experimental materials comprised male chickens of the Green-legged Partridge breed (old traditional Polish chicken breed), raised to 28 weeks of age. At 8 weeks of age, 100 birds were castrated. Caponization had a significant effect on the plasma concentrations of total cholesterol, high-density lipoprotein cholesterol, low-density lipoprotein cholesterol and triacylglycerols (P<0.05). Fatty degeneration and lymphoid cell infiltration were observed more frequently in the livers of capons than cockerels. Capon carcasses were characterized by increased deposition of abdominal and subcutaneous fat (P<0.05). Total meat weight in the carcasses of cockerels and capons was similar, but the proportions of muscles were different. From 20 weeks of age, the weight of breast muscles was higher, and the weight of leg muscles was lower in capons than in cockerels (P<0.05). Capons were characterized by higher liver weight, higher gizzard weight and lower heart weight than cockerels (P<0.05). The feed conversion ratio (kg/kg BW) was similar in intact cockerels and capons. The values of carcass quality parameters and feed conversion ratio as well as histopathological changes in the liver indicate that Green-legged Partridge capons should be slaughtered at 20 to 24 weeks of age.  相似文献
